This invention termed as Nanosystem for DNA on-the-spot tests (NaDOTS) offers an inexpensive supplementary test yet highly sensitive technique in addition to the gold standard to compensate the current RT-PCR protocol for a wider on-site sampling and frequent testing. NaDOTS consists of three modules namely sampling, amplification, and sensing. The sensing module is based on DNA biosensor technology. Electrochemical DNA biosensor is one of the most favoured devices due to its fast response and ultra-sensitivity. In DNA biosensors, the sensitive part consists of a single-stranded DNA (ssDNA) molecule which permits a hybridisation event with another complementary ssDNA molecule. The construction of electrochemical DNA biosensors comprises three major steps, which are the ssDNA probe modification, hybridization between target DNA and the probe DNA, and finally, the determination of signal output using specific detectors, which in this case, using electrochemical impedance spectroscopy (EIS).
